Presence of IgE Autoantibodies Against Eosinophil Peroxidase and Eosinophil Cationic Protein in Severe Chronic Spontaneous Urticaria and Atopic Dermatitis

PURPOSE Eosinophils are frequently found in atopic dermatitis (AD) and chronic spontaneous urticaria (CSU) that release eosinophil peroxidase (EPX) cationic protein (ECP). Continuous exposure to these proteins could trigger an autoimmune response which may contribute the pathogenesis severity of skin inflammation. In this study, we investigate immunoglobulin E (IgE) against CSU AD. METHODS We recruited patients with severe AD, healthy subjects explore presence IgE autoantibodies cross-reactivity EPX, ECP thyroid (TPO). The potential cross-reactive epitopes among family were determined using silico tools. RESULTS frequencies anti-EPX (28.8%) anti-ECP (26.6%) higher AD group, anti-TPO was group (27.2%). there a correlation between levels (r = 0.542, P < 0.001); TPO inhibited 42% binding while EPX 59% TPO, suggesting as primary sensitizer. There greater inhibition when used pool sera 52% 78% TPO. analysis showed possible shared epitope family. CONCLUSIONS inflammation CSU. Cross-reactivity explain problems patients.
